Abstract

Background: Angiogenesis is the process of developing new blood vessels from pre-existing vessels during the early stages of vasculogenesis. The purpose of this work was to synthesize, characterize the nanoformulated naringenin (NF-n) and evaluate the anti-angiogenic activity against native form on in ova chick chorioallantoic membrane (CAM) model.

Materials and methods: The nano solvent evaporation method was used to synthesize NF-n, and then it was characterized using various analyses. On the CAM model, the anti-angiogenic activity of nanoformulated naringenin was assessed by various parameters such as histopathology, immunohistochemistry and mRNA expression.

Results: Using the nano solvent evaporation method, NF-n was successfully synthesized and characterized by dynamic light scattering (DLS) and Fourier transform infrared spectroscopy (FT-IR). The morphological analysis of CAM treated with NF-n revealed a significant decrease in the blood vessels formation. The histopathological results of CAM treated with NF-n showed no significant changes in blood vessels. The immunohistochemistry results of CAM treated NF-n showed no significant expression of vimentin compared to control and native forms. Moreover, CAM treated with NF-n significantly decreased the mRNA expression levels of VEGF-A compared to control and native form.

 Conclusion: Taken together, results from present study suggest that NF-n functions inhibit the angiogenesis by stopping the activation of endothelial cells and down regulating angiogenic factors. Furthermore, the combinational treatment of NF-n and d provides better accuracy than individual treatment.
